A new architecture and operational techniques for supporting high quality live and on-demand streaming multimedia on a data network. By using Helper machines inside the network as data forwarding, caching, and buffering agents, and by forming meshes among Helper machines, advantages of homogeneous, synchronous multicast transmission and of heterogeneous, asynchronous reception are achieved. The architecture provides essentially transparently support to the receivers for near-zero start-up latency, improved playback quality, improved CR-like operations. One approach to treating individuals infected with HIV-1 is to administer to such individuals compounds that directly interfere with and intervene in the machinery by which HIV-1 replicates itself within human cells. Although the specific role of HIV-1 viral protein Vif in the viral life cycle is not known, the vif gene is essential for the pathogenic replication of lentiviruses in vivo. The present invention relates to a method for treating an individual exposed to or infected with HIV-1. Individuals identified as being exposed to or infected by HIV-1 are administered a therapeutically effective amount of one or more compounds that inhibit or prevent replication of said HIV-1 by interfering with the replicative or other essential functions of HIV-1 viral protein Vif, by interactively blocking the multimerization domain of Vif, thereby preventing multimerization of Vif protein, which is important for Vif function in the lentivirus life cycle. In preferred embodiments, the compound or compounds that interactively block the multimerization domain of Vif are Vif antagonists. Pharmaceutical compositions comprising these compounds are also disclosed.

Abstract:
The HIV-1 protein Vif comprises a multimerization domain that allows Vif-Vif interaction and Vif multimerization, which is important for Vif function in the HIV-1 life-cycle. A method for screening for an antagonist of Vif comprises contacting the multimerization domain of Vif with a test compound that specifically binds the multimerization domain. Antagonists identified by the screening assay inhibit Vif multimerization. The antagonists inhibit essential functions of Vif and accordingly are useful as inhibitors of HIV-1 replication.

Traditional networked file systems like NFS do not extend to wide-area due to network latency and dynamics introduced in the WAN environment. To address that problem, a wide-area networked file system is based on a traditional networked file system (NFS/CIFS) and extends to the WAN environment by introducing a file redirector infrastructure residing between the central file server and clients. The file redirector infrastructure is invisible to both the central server and clients so that the change to NFS is minimal. That minimizes the interruption to the existing file service when deploying WireFS on top of NFS. The system includes an architecture for an enterprise-wide read/write wide area network file system, protocols and data structures for metadata and data management in this system, algorithms for history based prefetching for access latency minimization in metadata operations, and a distributed randomized algorithm for the implementation of global LRU cache replacement scheme.
