Process For Selecting An Enantiomer Of A Hydroxy Lactone Using Pseudomonas Lipase

US Patent:
5413935, May 9, 1995
Filed:
Mar 24, 1993
Appl. No.:
8/106182
Inventors:
Ramesh N. Patel - Bridgewater NJ
Clyde G. McNamee - Lawrenceville NJ
Laszlo J. Szarka - East Brunswick NJ
Assignee:
E. R. Squibb & Sons, Inc. - Princeton NJ
International Classification:
C12P 4100
US Classification:
435280
Abstract:
An enantiomerically pure compound of the formula ##STR1## is prepared when the associated racemic mixture is reacted with an acrylating agent R. sup. 3 --C(O)--O--R in the presence of a microorganism or enzyme derived therefrom capable of catalyzing transesterification of an alcohol. X. sup. 1 and X. sup. 2 are each independently halogen, R is alkyl, R. sup. 1 and R. sup. 2 are each independently alkyl, cycloalkyl, aralkyl or aryl and R. sup. 3 is alkyl, cycloalkyl, aryl or aralkyl.

Enantioselective Preparation Of Halophenyl Alcohols And Acylates

US Patent:
5986095, Nov 16, 1999
Filed:
Jan 6, 1992
Appl. No.:
7/817232
Inventors:
Ronald L. Hanson - Morris Plains NJ
Ramesh N. Patel - Bridgewater NJ
Laszlo J. Szarka - East Brunswick NJ
Assignee:
E.R. Squibb & Sons, Inc. - Princeton NJ
International Classification:
C07D40304
US Classification:
544295
Abstract:
A process is described for preparing a compound of the formula or the S-enantiomer thereof, wherein: R is alkyl, aryl, cycloalkyl, aralkyl, or cycloalkylalkyl, R. sup. 1 is halogen; R. sup. 2 is halogen, alkyl, cycloalkyl, aryl or ##STR1## wherein the process comprises treating the associated racemic alcohol with an acylating agent ##STR2## (wherein L is a leaving group) and an enzyme or microorganism capable of enantioselective acylation. This process may also be used to isolate the unreacted R- or S-alcohol. The acylated product may be enantioselectively hydrolyzed with a lipase or lipase-supplying microorganism to the S- or R-alcohol. Compounds prepared by this invention are useful antipsychotic agents or useful intermediates therefor.
