Methods are provided for identifying agents that modulate the expression of genes, such as genes encoding tumor suppressor or tumor-promoting proteins. This invention provides a method for preparing antibodies directed against a protein associated with a cancer, comprising the steps of: (a) transfecting non-tumorigenic cells with a cDNA library comprising cDNA molecules derived from mRNA of cancer cells; (b) masking the transfected cells with antibodies raised against non-tumorigenic cells transfected with a cDNA library derived from normal cells; and (c) generating antibodies against the masked cells, and thereby generating antibodies directed against a protein associated with a cancer.
