Caged Enzyme Substrates As Probes For Reporter Enzyme Activity

US Patent:
5981207, Nov 9, 1999
Filed:
Dec 18, 1998
Appl. No.:
9/216445
Inventors:
Jonathan J. Burbaum - Cranbury NJ
Ke Yang - West Windsor NJ
Assignee:
Pharmacopeia, Inc. - Princeton NJ
International Classification:
C12Q 142
C12Q 100
US Classification:
435 21
Abstract:
Caged enzyme substrates as probes for detecting reporter enzyme activity in cell-based assays are disclosed. Caged substrates for reporter gene assays are shown as compounds of Formula I or Formula II: ##STR1## wherein Z is a luminescent functionality; G is a labile group, cleavable by enzymatic action; and K is a photolytically cleavable caging group; wherein m and n are independently 0 or 1, but m and n both can not be zero. Test kits for determining reporter enzyme activity using compounds of Formula I or Formula II and photolytic cleavage to produce a compound of Formula III: G. sub. m --Z--G. sub. n III are described. Methods for determining reporter enzyme activity and, by inference, the activator or suppressor activity of a test compound in a cell-based assay are also described.

Rotary Actuator Utilizing Pneumatically Actuated Elastomeric Structures

US Patent:
2017014, May 25, 2017
Filed:
Nov 18, 2016
Appl. No.:
15/355861
Inventors:
- New Brunswick NJ, US
Ke Yang - Parlin NJ, US
Xiangyu Gong - Edison NJ, US
International Classification:
F01B 19/04
F15B 15/10
Abstract:
A rotary actuator and a method of using same are disclosed. The rotary actuator includes a rotor having a body and defining a plurality of contact surfaces, and a stator having a body and defining a plurality of inflatable bladders circumferentially spaced about the stator body. The stator is positioned relative to the rotor such that upon sequential inflation of the plurality of inflatable bladders, the rotor is caused to rotate.
